OPTIMASI DAN KOMBINASI FORMULA PERTUMBUHAN LACTOBACILLUS CASEI MENGGUNAKAN MOLASE DAN SUSU SKIM

Abstract

Medium pertumbuhan bakteri merupakan suatu bahan yang terdiri dari nutrisi yang diperkaya dengan bahan tertentu untuk pertumbuhan bakteri, isolasi, memperbanyak jumlah, menguji sifat-sifat fisiologi dan perhitungan jumlah mikroba. Medium terbaik buat pertumbuhan bakteri adalah sesuai dengan unsur-unsur pertumbuhan bakteri harus mengandung sumber karbon, mineral, vitamin dan gas, derajat keasaman (pH).

Susu Skim dapat mempengaruhi pertumbuhan bakteri asam laktat. Semakin tinggi persentase susu skim akan dapat mempengaruhi pertumbuhan bakteri. Pada susu skim banyak terdapat kandungan laktosa yang bermanfaat untuk pertumbuhan bakteri.

Penelitian ini merupakan uji eksploratif yang bertujuan untuk menentukan formulasi dan optimalisasi medium pertumbuhan bakteri Lactobacillus casei dengan kombinasi molase dan susu skim. Pengukuran pertumbuhan Lactobacillus casei menggunakan pengukuran cahaya yang ditransmisikan pada panjang gelombang 400-600nm.

Dari hasil penelitian pada keempat media pertumbuhan yaitu Media A (Molase 1,5% Susu skim 5%) didapatkan hasil 846.08 NTU, Media B (Molase 1,5%, Susu skim 2,5%) didapatkan hasil 851.32 NTU, Media C (Molase 3%, Susu skim 5%) didapatkan hasil 880.30 NTU, Media D (Molase 3%, Susu skim 2,5%) 852.30 NTU. Dapat disimpulkan bahwa Media C (Molase 3%, Susu skim 5%) merupakan media yang  paling optimal dari pada semua media sebagai formula pertumbuhan bakteri Lactobacillus casei.
